P-73: Effect of Donor Age on The Expression Stability of GAPDH as A ReferenceGene for Gene Expression Analysis ofEquine Adipose-Derived Mesenchymal Stem Cells
Authors
Abstract:
Background: Adipose tissue is a main source for isolation of equine mesenchymal stem cells (MSCs) at different ages. It seems that characteristics of adipose-derived MSCs especially gene expression profile are changing along with age increase. A proper reference gene is required for normalizing data in gene expression analysis by qRT-PCR. This study aimed to evaluate whether GAPDH has a stable expression in adipose-derived MSCs isolated from horses with different ages. Materials and Methods: MSCs isolated from adipose tissue of 3 mares, aged 3, 6 and 9 years old. After 3 passages, total RNA of cultured cells was isolated and mRNA reverse transcribed to cDNA. qRT-PCR was done using GAPDH primers in triplicate. Results: Our analysis indicated that Ct of GAPDH gene is very similar in all samples. Thus, its expression was almost the same at adipose-derived MSCs of horses with different ages. Mean ± SD of Ct was 15.53 ± 0.47. Conclusion: Based on the results, it suggests that expression level of GAPDH is stable in MSCs of horses with different ages. Thus, it can be considered as a suitable reference gene for gene expression analysis for adipose-derived MSCs isolated at different ages.

Comparative analysis of the Gene expression profile of Chemokine Receptors between Adipose-derived and Bone marrow-derived Mesenchymal Stem Cells
Introduction: Mesenchymal stem cells (MSCs) hold great promise in the field of regenerative medicine.Although originally isolated from bone marrow, MSCs have since been obtained from a variety of adult and neonatal tissues including the adipose tissue. Stemness and multipotential features of Mesenchymal Stem Cells (MSC) has been highlighted in many studies but there are many dark aspects in ex...
full textEvaluation of β-actin as a Reference Gene for Comparative Expression Analysis of Equine Adipose- and Bone Marrow-Derived Mesenchymal Stem Cells by qRT-PCR
Background Bone marrow and adipose tissue are two main sources of mesenchymal stem cells (MSCs). Some of studies suggest that there are some differences in gene expression profile of MSCs-derived from various tissues. To investigate gene expression profile by qRT-PCR, an appropriate reference gene with stable expression level should be chosen for normalizing data. This study was designed to e...
full textGene Expression Profile of Adherent Cells Derived From Human Peripheral Blood: Evidence of Mesenchymal Stem Cells
Mesenchymal stem cells (MSCs) provide a novel option in cellular therapy and tissue engineering. Recent studies indicated that it is possible to obtain MSCs from peripheral blood by attachment ability to plastic surface. To evaluate adherent cells derived from peripheral blood, their expression profile and surface markers were investigated. The results of RT-PCR indicated that these cells expre...
full textDifferential gene expression by lithium chloride induction of adipose-derived stem cells into neural phenotype cells
Objective(s): Adipose-derived stem cells (ADSCs), with suitable and easy access, are multipotential cells that have the ability for differentiation into other mesodermal and transdifferentiate into neural phenotype cells. In this study, Lithium chloride (LiCl) was used for in vitro transdifferentiation of rat ADSCs into neuron-like cells (NLCs).<stro...

full textPlatelet-derived Microparticles increase the Expression of hTERT Gene in Umbilical Cord Mesenchymal Stem Cells
Background: Mesenchymal stem cells have been widely considered in clinical researches because of their self-renewality and differentiation into various tissues. Nevertheless, their limited in vitro life span, which occurs only after several divisions, makes some changes in these cells, which affects all of their characteristics and remarkably reduces their application. In this study, the effect...
